Start with Stage 2 and walk through the first area and into the building. Once you enter, you'll come across some locked doors with someone shouting, "Help!" Attack the door to open it up, and Barbara will come running out from the cell. You can use Stage Select to access Stage 2 directly from the Character Select screen.
